http_port 80 defaultsite=your.main.website vhost

you most likely do not want vport.

and cache_peer + cache_peer_access/domain to route the requests to the
correct server, alternatively always_direct if you want to route
by /etc/hosts like before..

nope, not for accelerators. This is for transparently intercepting
proxies.
