No. You can configure big cache_mem to get better memory hit ratio and to
avoid some disk I/O, which is probably the most frequent bottleneck of squid
caches.

You can increase maximum_object_size_in_memory when you increase cache_mem.
with 4GB of RAM the 64MB meomry cache is quite low and you can increase it,
unless you have very big disk cache and the squid process eats 2 GB of RAM
also without the memory cache. That should happen if you have >~10 millions
of object in the cache.
