Does your "cache_effective_user" have write permission to
/usr/local/squid/var/?
Try
mkdir /usr/local/squid/var/cache
chown your_cache_effective_user /usr/local/squid/var/cache
/usr/local/squid/sbin/squid -z -d3
/usr/local/squid/sbin/squid -DNYCd3
