If you need to use async-io then please upgrade to Squid-2.4 or
downgrade to Squid-2.2.STABLE5-hno. async-io in Squid-2.3 has a fair bit
of issues.
To answer your question: No you do not need to clean your cache. The
on-disk format is identical.
